Issues in drug delivery arebecoming more important as more potent and specific drugs become available with the knowledge about diseases available from the human genome project. To achieve optimal therapeutic efficacy, drug delivery systems should be customized very innovative. Novel systems are often nanosized, prepared by specific excipients, and they have multifaceted advantages in drug delivery. These systems in general can be used to provide targeted (cellularžtissue) delivery of drugs, to improve oral bioavailability, to sustain drugžgene effect at target place, and to improve the stability of therapeutics agents against enzymatic degradation, especially of protein, peptide and nucleic aciddrugs. Vectors for gene delivery could be divided into two major groupsČ viral and nonviral. Due to several limitations of viral vectors more attentions attract nonviral vectors, which consist of a carrier with cationic character able to associate with negatively charged DNA molecule. Formation ofthe complex protects DNA against denaturation and enables its entry into thecells. For specific processes that must take place, such as cell recognition, endocytosis, cytosolic trafficking, endosomal escape, dissociation of complex, efficient nuclear uptake and gene expression, severalspecific molecules are attached into gene delivery devices enabling targeting, fusion with the membrane, changes in the microenviroment or in other way affects cellular function for successful transgenic expression. Combinations of viral and nonviral components are hybrid vectors, which are safer as viral and more efficient as nonviral vectors"},{"@xml:lang":"sl","#text":"V prispevku predstavljamo sisteme in moznosti vnosa proteinskih ueinkovin in genov v telo. Dostavljanje ueinkovin na mesto delovanja postaja vse bolj pomembno, saj so se z odkritjem genoma in s tem povezanim znanjem o gensko zasnovanih boleznih pojavile ueinkovitejse in zelo specifiene ueinkovine. Novisistemi so poveeini nanometrskih velikosti (nanodelci, liposomi, nanokompleksi), zgrajeni iz doloeenih nosilnih snovi, in imajo sirok spekter prednosti. Omogoeijo ciljanje v tkivožcelico, poveeajo biolosko uporabnost ueinkovine, ji podaljsajo delovanje na ciljnem mestu ter nudijo zaseito pred encimsko razgradnjo, se posebej proteinom in nukleinskim kislinam. Vektorji zavnos genskega materiala v celico so virusni in nevirusni. Zaradi velikih pomanjkljivosti virusnih vektorjev postajajo zanimivejsi nevirusni vektorji, ki vkljueujejo nosilec s kationskim znaeajem, kar omogoea povezovanje z negativno nabito molekulo DNA. Tvorba kompleksov pomeni zaseito genskega materiala in olajsa vnos v celico. Za vodenje procesov v celici, kot so endocitoza, izstop iz lizosoma, razpad kompleksa, vstop v jedro in vkljueevanje v jedrno DNA, pripnejo na nosilni sistem specifiene molekule, ki poveeajo ciljanje, zlitje z membranami, spremenijo mikrookolje v celici ali drugaee vplivajo na celiene funkcije, ki olajsajo vnos genskega materiala v jedro.
