These materials include biologically active proteins, probiotics, polymorphic drugs and some excipients. Therefore tableting of these materials is performed at low pressures. Traditional excipients like microcrystalline cellulose, hyroxypropylmethyl cellulose and dicalcium phosphate dihiydrate are often unable to form stable and robust tablets at lowpressure. Excipients with greater elastic deformation are more suitable forsoft tableting. From a theoretical point of view, mechanical properties of materials are best described by parameter of general plasticity (GP). This parameter includes parameters describing materials time plasticity, pressure plasticity, elastic decompression and elastic recovery. The lower the GP of material, more suitable it is for tableting at low pressure. The correlation between GP and stability of pressure-sensitive material was found out. Excipients with low GP can reduce the transformation among different drug polymorphic forms due to tableting, they are able to prevent inactivation of incorporated enzymes, and can prevent damages of functionally coated pellets during tableting. According to literature data the most suitable excipients for soft tableting are the carrageenans</dc:description><dc:description xml:lang="sl">Vse več je snovi, ki so občutljive na visoke tlake med tabletiranjem. Sem sodijo različni biološko aktivni proteini, probiotične snovi, polimorfne zdravilne učinkovine in nekatere pomo-ne snovi. Zato tabletiranje teh snovi izvajamo pri relativno nizkih tlakih.Tradicionalna polnila, kot so mikrokristalna celuloza, hidroksipropilmetilceluloza in dikalcijev fosfat dihidrat pri nizkih tlakih pogosto ne omogočajo izdelave stabilnih in robustnih tablet. Za t. i. mehko tabletiranje (angl. soft tableting) so tako primernejše snovi, ki imajo bolj elastične lastnosti. Mehanske lastnosti snovimed tabletiranjem in po njem teoretično najbolje opisuje parameter splošne plastičnosti (angl. general plasticity, GP). Slednji vključuje parametre, ki opisujejo lastnosti materialov v smislu njihove časovno odvisne plastičnosti; plastičnosti, ki je odvisna od tlaka; elastičnosti materiala in indeks elastične relaksacije. Nižja kot je GP, bolj primerna je snov za tabletiranje pri nizkih tlakih. Ugotovljena je bila direktna korelacija med GPin stabilnostjo snovi, občutljivih na povišan tlak. Pomožne snovi z nizko GPlahko zmanjšajo obseg prehajanja med polimorfnimi modifikacijami učinkovin med tabletiranjem, zagotavljajo ohranjanje aktivnosti vgrajenih encimov in preprečujejo poškodbe funkcionalne obloge pelet med tabletiranjem.
