Its inhibition presents a promising target in cancer with the option of combinatorial attack on numerous oncogenic pathways. Clinical Hsp90 inhibitors bind to N-terminal ATP-binding pockets, suppressing ATPase activity and exhibiting pan-inhibitory activity. Adverse effects of said inhibition that include heat shock response lead to the development of new strategies for Hsp90 inhibition. Novel Hsp90 inhibitors offer selective suppression of Hsp90 isoforms or specific disruption of Hsp90-co-chaperone protein-protein interaction, C-terminal domain inhibition and inhibition of Hsp90 post-translational modifications. While they have yet to enter clinical trials their results from preclinical stages show a great potential for future research of Hsp90 inhibitors</dc:description><dc:description xml:lang="sl">Hsp90 je eden najpomembnejših šaperonov, vključenih v regulacijo celične homeostaze ter dozorevanje in zvitje številnih proteinov, med katerimi so mnogi povezani z napredovanjem raka in ostalih – predvsem nevrodegenerativnih – bolezni. Ker lahko z njegovo inhibicijo hkrati vplivamo na več onkogenih poti v celici, predstavlja zanimivo tarčo v terapiji raka. Zaviralci Hsp90, ki so bili do sedaj v kliničnih študijah, se vežejo v ATP-vezavno mesto N-končne domene in neselektivno zavirajo ATPazno aktivnost vseh štirih izoform. Neželeni učinki take inhibicije, med katerimi je tudi odziv toplotnega šoka, so bili povod za odkritje alternativnih poti inhibicije Hsp90. Novejši inhibitorji tako izkazujejo izoformno selektivno delovanje, vplivajo na interakcije protein–protein med Hsp90 in njegovimi košaperoni, delujejo na C-končno domeno in se vpletajo v posttranslacijske modifikacije.
