Development of endometrial cancer is usually explained with the unopposed estrogen hypothesis, which proposes that exposure to estrogens that is not opposed by progestagens stimulates proliferation of endometrium and increases DNA replication errors. This may lead to endometrial hyperplasia and cancer. Studies from the last years suggest that also androgens are implicated. Epidemiological data show associations between the increased serum levels of androgens and increased risk of endometrial cancer, although studies on cell lines and cancerous tissue reveal their protective role.
