Namen metode je napraviti vidne specifične beljakovinske molekularne komplekse v celicah ali v medceličnini. Za dokazovanje tarčnih molekul se uporablja primarna protitelesa, ki se specifično vežejo le na iskane molekule. Molekulske komplekse nato označimo s sekundarnimi protitelesi, ki imajo vezane označevalce. Označevalci omogočajo opazovanje in kvantifikacijo kompleksov z mikroskopom.
